Do I transfer my newly purchased domain name (.com.au) before or after I sign up with Blue Voda?

You won't transfer it, you will just change the nameservers to the ones that will be sent in the welcome email. Country specific domain name, like .com.au cannot be registered with company's outside of Australia.
